The filling process begins with the manual removal of the Tyvek sealing paper and protective plastic lid from the sterile syringe tub. The operator carefully opens the ready-to-use (RTU) syringe tub while following standard aseptic handling procedures to maintain the sterility of the syringe nest. Since the syringes have already been sterilized by the manufacturer, proper handling during this stage is essential to prevent contamination before the filling operation begins.

The Tyvek membrane is peeled away smoothly using controlled manual handling to avoid generating particles or damaging the sterile packaging. After the Tyvek seal is removed, the protective plastic lid is lifted carefully to expose the syringe nest. The operator ensures that the nest remains properly seated inside the tub and that no syringes are displaced during the opening process. This careful handling preserves the integrity of the pharmaceutical packaging and prepares the syringes for automated processing.

Before the syringe tub is transferred to the filling station, the operator performs a visual inspection to confirm that the Tyvek membrane and lid have been completely removed and that the syringe nest is clean, correctly positioned, and free from visible damage or foreign particles. Any abnormality is identified before processing continues, ensuring that only properly prepared syringe tubs enter the filling cycle.

Although this stage is performed manually, it plays a critical role in maintaining aseptic conditions and ensuring reliable downstream operations. After opening the syringe tub, the operator manually places it onto the machine’s filling station. Precision guide rails and locating pins assist in positioning the tub accurately, ensuring that the syringe nest is aligned correctly before the automatic cycle begins. Proper placement is essential for achieving consistent nest transfer and precise filling performance.

Once the tub is positioned, the machine’s sensors verify its presence and confirm correct alignment. The PLC control system checks that the tub is securely seated before allowing the production cycle to start. If the tub is not positioned correctly, the machine alerts the operator through the Human Machine Interface (HMI), preventing incorrect processing and protecting both the syringes and the machine.

The stable positioning system prevents movement of the syringe tub during automated handling. By securely holding the tub in place, the machine ensures that the syringe nest can be transferred safely without disturbing the sterile syringes. Accurate positioning also improves filling precision by maintaining consistent alignment between the syringe nest and the filling table.

This combination of manual loading and automatic position verification provides an efficient balance between operator control and machine automation. It minimizes setup errors while maintaining high levels of repeatability, productivity, and pharmaceutical quality.

Nest Transfer to Filling Table

Nest Transfer to Filling Table : – 

Once the syringe tub has been verified, the machine automatically transfers the syringe nest from the tub to the precision filling table. A servo-controlled lifting mechanism gently raises the nest while maintaining its original orientation. The transfer motion is smooth and vibration-free, preventing damage to the syringes and preserving their sterile condition.

The filling table is equipped with precision locating mechanisms that position the syringe nest with exceptional accuracy beneath the filling nozzles. This precise alignment ensures that each filling nozzle enters the corresponding syringe correctly, enabling highly accurate liquid dosing and consistent pharmaceutical quality.

Throughout the transfer process, integrated sensors continuously monitor the position of the syringe nest. The control system confirms successful transfer and correct placement before initiating the filling operation. If any positioning error is detected, the machine automatically stops and alerts the operator, preventing processing faults and protecting the product.

The automated nest transfer system significantly reduces manual handling of sterile syringes, lowering contamination risks while improving operational efficiency. Its precise and repeatable motion contributes to reliable filling performance and consistent production quality.

Simultaneous Filling and Stoppering

Simultaneous Filling and Stoppering : – 

After the syringe nest is accurately positioned on the filling table, the machine begins the simultaneous filling and stoppering operation. High-precision filling nozzles dispense the programmed volume of pharmaceutical liquid into each syringe using servo-driven dosing technology. The filling process is designed to provide excellent accuracy while minimizing product loss, foaming, and air entrapment.

Immediately after filling, rubber stoppers are automatically fed through an orientation system and inserted into each syringe. The stoppering mechanism applies controlled insertion force to ensure secure sealing without damaging the syringe components. Performing filling and stoppering in one synchronized operation reduces production time while maintaining aseptic processing conditions.

During this stage, the PLC continuously monitors critical process parameters such as fill volume, nozzle position, filling pressure, and stopper placement. Integrated sensors verify that every syringe has been correctly filled and sealed before allowing the production cycle to continue. Any abnormal condition automatically generates an alarm, ensuring high product quality and process reliability.

Return of Filled Nest to Tub

Return of Filled Nest to Tub: – 

After filling and stoppering have been successfully completed, the machine automatically transfers the filled syringe nest back into its original tub. The servo-controlled transfer mechanism gently lifts the completed nest from the filling table and positions it precisely inside the tub without disturbing the filled syringes. Smooth motion and accurate positioning help maintain product integrity throughout the handling process.

Precision guides ensure that the syringe nest is seated correctly inside the tub before the transfer cycle is completed. Sensors verify successful placement and confirm that the filled nest is properly aligned for downstream handling. This automated verification reduces handling errors while ensuring consistent production quality.

Returning the filled nest to the original tub protects the syringes during transportation to subsequent pharmaceutical operations such as inspection, labeling, secondary packaging, or cold storage. By minimizing manual handling after filling, the machine further reduces the risk of contamination and mechanical damage.

The automatic nest return system completes the semi-automatic production cycle with high precision and reliability. By combining controlled motion, intelligent monitoring, and GMP-compliant hygienic design, it ensures that every filled syringe nest is safely prepared for the next stage of pharmaceutical manufacturing while maintaining sterility, product quality, and operational efficiency.
